Hatcher made her motion picture debut in Christopher Guest's 1988 Hollywood satire The Big Picture, starring Kevin Bacon. She then appeared with Sally Field, Kevin Kline, Whoopi Goldberg and Robert Downey Jr. in the hit comedy Soapdish, and with Dolly Parton and James Woods in Straight Talk. She went on to star in the 1996 erotic thriller Heaven's Prisoners, opposite Alec Baldwin and Eric Roberts, and in 2 Days In the Valley, with James Spader, Jeff Daniels, Danny Aiello, and Eric Stoltz.

In 1997, Hatcher joined the legacy of the popular James Bond film series when she co-starred with Pierce Brosnan and Jonathan Pryce in Tomorrow Never Dies. She also appeared in Alex Winters' Fever, an independent thriller with Henry Thomas, and the hit action-comedy Spy Kids, with Antonio Banderas, Carla Gugino, and Alan Cummings.

On television, Hatcher starred in Norman Lear's semi-autobiographical series Sunday Dinner, and starred for four seasons as Lois Lane in the hit ABC series Lois & Clark: The New Adventures of Superman. She has also made guest-starring appearances on numerous series including Seinfeld, L.A. Law, Murphy Brown, and Frasier, and hosted NBC's Saturday Night Live. She was also seen in the TNT original movie Running Mates with Tom Selleck, Laura Linney, Nancy Travis, and Faye Dunaway. In 1996, Hatcher made her debut as a screenwriter with an episode of Lois & Clark entitled It's a Small World After All.

In 2000, Hatcher made her stage debut, starring as Sally Bowles in the national tour of the Tony Award-winning musical Cabaret. She then returned to the stage in Eve Ensler's acclaimed The Vagina Monologues, performing in both the Off-Broadway and Los Angeles productions.
